You bascially have to know what you are wanting to get and how it's being done beforehand for some of the stores. Hasbro, for instance, hands out tickets early in the morning with a time when you can go to the booth. The first day of the Con a lot of people try to get all their exclusives so it can be a battle with long lines and such because no one wants to go latter to find it's sold out or miss a panel they really want on another day.

A lot of them have daily limits and sometimes it's just a free for all once the doors open. Last year my buddy wanted a Portal Gun so I got in first and went to the line. Little did I know the line I was in was for something else and the Portal Guns were right there next to me with no line out of what looked like a back door. By the time someone explained their were more people in the Portal Gun line than the daily allotment. I was pretty angry because I was staring the guy in the face for 3 minutes and he never said anything nor was there any sign posted. That is why you gotta figure out the process for what you want beforehand lol.
