Locke was referring to the episode, where the old couple were poisoning people and feeding them to the "Horror" tree in front of their house. They were doing their same routine, but was shocked to see that it didn't work on Zero.

Their suits protect them when they are actually wearing them. But even then they can still be harmed, if it's a powerful enough enemy. If what you say was even remotely true. Then "Red Mask" would've been able to put that "Death Mark" on every last Makai Knight.

Just keep watching Locke. Men are the only ones who can become Makai Knights. Because Men are physically stronger then Women. So that is why the women become Makai Priestess and the men either a Makai Knight or Makai Priest.
